James Hamilton was a great2-grandson of James and Janet (Hamilton) Muirhead.(3.32) He descended through their daughter, Margaret, who married John Hamilton, 6th of Udstoun. Their son, James Hamilton 1st of Barncleuch, married Margaret Hamilton, and their son, Robert married Marion Denholm. Their second eldest son was James 1st of Pencaitland.
James Hamilton, son of Robert and Marion Hamilton, married Catherine Denholm. Catherine was a daughter of James Denholm of Weshiels. James was given the title of 1st of Pencaitland, Senator. The couple gave birth to: John; Alexander; Marion, baptised 17 February 1685; Robert, baptised 16 July 1686; Anna, baptised 08 August 1687; and Margaret. James died in the year 1729.
John Hamilton, son of James and Catherine Hamilton, married Margaret Menzies. Margaret was a daughter of Alexander Menzies of Saltcoats. They gave birth to: James, born 25 January 1710. The child died young. John held the title of 2nd of Pencaitland.

Anna Hamilton, daughter of James and Catherine Hamilton, married Sir Archibald Grant 2nd Bart of Monymusk, on 17 April 1717. Archibald was born in the year 1696 and died on 17 September 1778.
Margaret Hamilton, daughter of James and Catherine Hamilton, married Sir Alexander Gibson of Pentland, 3rd Bart. Alexander died on 15 April 1774.
